NAP (NAPVSIPQ) is an 8-amino acid peptide derived from activity-dependent neuroprotective protein (ADNP). It stabilizes microtubules, reduces tau hyperphosphorylation, and provides potent neuroprotection at femtomolar concentrations. Phase II trials in Alzheimer's and schizophrenia showed cognitive improvements via intranasal delivery. Among the most potent neuroprotective sequences known.
Nasal irritation
Headache (uncommon)
Generally excellent safety profile at research doses
Assuming more is better — NAP is effective at femtomolar concentrations; Phase II trials used 5–30 mcg; exceeding this doesn't improve outcomes and may reduce selectivity
Not using intranasal delivery — oral NAP does not meaningfully reach the CNS; intranasal is the only route with clinical evidence
Expecting immediate cognitive changes — neuroprotection is cumulative over months; short-term cognition enhancement is not NAP's primary mechanism
Cerebrolysin — complementary neuroprotection; tau stabilization (NAP) + neurotrophic support (Cerebrolysin) for comprehensive CNS coverage
Semax — synergistic; microtubule stabilization (NAP) + BDNF/NGF upregulation (Semax) for the most evidence-based nootropic pairing
No known significant adverse drug interactions at research doses
NAP is extraordinary in its potency — effective at doses 1,000× lower than most research peptides. The tau-stabilizing and microtubule-protective effects make it the most specific Alzheimer's-preventive peptide available today. 30 mcg intranasal daily + Semax 300 mcg is the best neuroprotection stack in this entire index for long-term cognitive preservation.
